2、bruary1,1979Ahumancoronavirus,strainOC43,waspropagatedinsucklingmousebrainandpurified5000-foldwitha90%yield.Purityoftheviruswasconfirmedbyelectropho-retie,ultracentrifugal,andelectronmicroscopicprocedures.Immunodiffusionandimmunoelectrophoresistestsrevealedoneprecipitinlinewithnor
3、malmousebrain,threewithpurifiedvirus,andfourwithcrudeviruswhentestedagainstanti-purevi-rusoranti-crudevirusanimalserums.TheassociationofahostcellantigenwiththevirionwasconfirmedbystandardHIandCFtests.Polyacrylamidegelelectrophoresisofsolubilizedpurifiedvirusrevealedaminimumofsixpo
4、lypeptideswithapparentmolecularweightsof191,000(No.I),104,000(No.2),60,000(No.3),47,000(No.4),30,000(No.5),and15,000daltons(No.6).Aseventhbandwasoccasionallyfoundinthe165,000-daltonregionofthegels.Fourpolypeptidescontainedcarbohydrateandonecontainedlipid.PolypeptideNo.5comprised26
5、7’ofthetotalviralproteinandglycopolypeptideNo.3comprised23%.Threeothercomponentsaccountedformostoftheremainingprotein:polypeptideNo.4(lG’%),glycopolypeptideNo.6(14%),andglycolipopolypeptideNo.1(13%).GlycopolypeptideNo.2was8%ofthetotalprotein.Bromelindigestionoftheviralprojections(
6、spikes)removedglycopolypep-tidesNo.2andNo.6.Associationoftheremainingpolypeptideswithstructuralcomponentsofthevirionisonlytentativelypostulated.Thebuoyantdensityinpotassiumtartrateofthebromelin-treatedviruswas1.15g/cm3andoftheintactOC43virionwas1.18g/ems.Byanalyticalultracentrifug
