2、978)SUMMARYAnalysisbySDS-polyacrylamidegelelectrophoresisshowsthatthepurifiedcoronavirusJHMcontainssixpolypeptides.Theapparentmol.wt.ofthepolypep-tides(GPr,GP2,GP3,VP4,GP5andVP6)are170ooo;125000;975oo;60800;24800and22700,respectively.Fourpolypeptidesaregly
3、cosylated(GPI,GP2,GP3andGP5).Theanalysisofparticlesobtainedafterlimitedproteolysiswithpro-nasesuggeststhatGP2andGP3areprotrudingfromthelipidenvelopeand,to-getherwithGPI,formthespikelayer.ProteinVP6andapartofGP5arelocatedwithinthelipidbilayer.ProteinVP4issu
4、sceptibletodigestionataconcentrationofpronasewhichchangesthemorphologyofthevirusparticlesmakingtheinteriorofthevirusaccessible.Subviralparticlesproducedaftertreatmentwiththedeter-gentNonidetP4obandedatahigherdensitythanthevirusandcontainedonlyVP4,GP5andVP6
5、.INTRODUCTIONInmanyanimalspecies,includingman,coronavirusesareassociatedwithagreatvarietyofdiseases(McIntosh,1974;Tyrrelletal.1975).Inmanycasesthevirusesofthisgroupareonlycharacterizedmorphologicallyandserologically.Duetothelackofsuitabletissueculturesyste
6、msforviruspropagationfewreportsonvirusstructureandreplicationofcoro-navirusesareavailable.Thepublisheddataonstructuralproteinsofthesevirusesare,insomecases,inconsistent(Hierholzeretal.i972;Bingham,1975;Garwes&Pocock,I975;Collinsetal.i976;Hierholzer,I976;Ma
7、cNaughton&Madge,1977;Sturman,1977).ThenumberofpolypeptidesreportedforcoronavirusesvariesfromI6proteins(Bingham,I975)foravianinfectiousbronchitisvirus(IBV)to4proteinsforthemousehepatitisvirusstrainA59(Sturman,1977).Manyofthesedifferencesmaybeaccountedforbyt
