3、s-specificprobes:cDNA~ep,representingthemajorityofsequencesofthegenomeRNAandcDNAy,representingthe3'endofthegenomeRNA.MolecularhybridizationwiththesecDNAswasusedtocharacterizebothgenomeRNAandintracellularvirus-specificRNAs.HybridizationofgenomeRNAsofMHVstrainsA59,JHM,andMH
4、V-3withA59cDNArepshowedthat,althoughthesethreestrainsexhibitdifferentpathogenicities,theycontaincloselyrelatednucleotidesequences.HybridizationofintracellularRNAfromMHV-infectedcellswithvirus-specificcDNAshowsthat(i)themajorityofvirus-specificRNAispolyadenylated,(ii)virus
5、-specificintracellularRNAcontainssixsubgenomicspeciesofthesamepolarityasgenomeRNAand(iii)allsubgenomicRNAscontainthesameYsequencesasthegenomeRNAandthusformanestedsetofRNAs.INTRODUCTIONCoronavirusescauseacuteand/orpersistentdiseaseinmanyspeciesofanimals.Onefactorthatdeterm
6、inesthetargetorganandlethalityofcoronavirusinfectionisthestrainofvirus(McIntosh,1974).Forexample,moststrainsofmousehepatitisvirus(MHV),suchasMHV-3,wereisolatedfromtheliversofinfectedmiceandareprimarilyhepatotropic(McIntosh,1974).TheA59strain,whichiscommonlyusedinthelabora
7、torybecauseitgrowstohightitresincellculture,isonlyweaklypathogenic(Robb&Bond,1979).TheJHMstrainisneurotropicandcausespersistentdemyelinatingdisease(Baileyetal.,1949).Infectionofrodentswiththisstrainhasbeencitedasamodelformultiplesclerosis.MostMHVstrainsarecloselyrelatedse
